Primary (1) put the normal intestinal tissue of mice after sampling into the sampling bottle of pre cooled (2-8° c) tissue preservation solution e, and quickly transfer it to the clean laboratory for tissue processing and stem cell separation, take photos and register information (2) prepare several Petri dishes and add 4 ℃ pre cooled primary culture buffer B for standby (3) disinfect the sampling bottle, put the tissue into the Petri dish, wash it three times with primary culture buffer B, remove impurities, remove the intestinal mucosa of small intestine tissue with ophthalmic scissors or scalpel, and cut it into a tissue block with a volume of about 1-3mm3 (4) the tissue was digested with the primary tissue digestive solution c of mouse normal intestine, and then it was shaken at 4 ℃ for 10min (the digestion condition was observed at any time during the digestion process) (5) take a small amount of liquid and observe it under the microscope. After observing more single cells or cell clusters below 70UM under the microscope, add three times the volume of primary culture buffer B to stop digestion (6) filter with a 100um pore size screen, collect the filtrate, remove the supernatant after centrifugation at 300g for 5min, add primary culture buffer B and resuspend for centrifugation (7) Matrigel calculation: after step 6, observe the collected tissue volume, add 25 times the tissue volume of Matrigel (abs9495) to resuspend the plate (8) take a 24 well cell culture plate as an example, dispense 25ul of tissue Matrigel mixture per well for plating (operate at 4 ℃) (9) put the laid culture plate into a 37 ℃ incubator for 10-15min to form a glue, and add mouse normal intestinal organoid medium a (restore room temperature) for culture 2. Organoid subculture (1) pipette the culture medium, add 1-2ml of 4 ℃ organoid subculture buffer g to each well and place it for 2min (2) pipette the Matrigel gently, collect it in a 15ml centrifuge tube, and let it stand at 4 ℃ for 10min. (each 6-8 well is a group) (3) a: the number of organoids is insufficient or the volume is small: centrifuge for 5min to discard the supernatant, add an appropriate amount of organoid subculture buffer g to resuspend and transfer into a 1.5ml centrifuge tube, centrifuge for 5min at 300g to discard the liquid for step 4  &Nbsp&Nbsp&Nbsp b: When the number of organoids is large or the volume is large: centrifuge for 5min to discard the supernatant, add an appropriate amount of organoid passage digest D for digestion for 2-3min, add organoid passage culture buffer g to stop digestion, centrifuge for 5min to discard the mixture, add an appropriate amount of organoid passage culture buffer g to resuspend and transfer into a 1.5ml centrifuge tube, centrifuge for 5min at 300g to discard the liquid for step 4 (4) after organ collection, add Matrigel to resuspend, lay 25ul Matrigel per well in a 24 well cell culture plate, place in the incubator for 10-15min, and add 500ul of mouse normal intestinal organoid culture medium a 3. Organoid cryopreservation (1) pipette the culture medium, add 1-2ml of 4 ℃ organoid subculture buffer g to each well and place it for 2min (2) pipette the Matrigel gently, collect it in a 15ml centrifuge tube, and let it stand at 4 ℃ for 10min. (each 6-8 well is a group) (3) centrifuge for 5min to discard the supernatant, add an appropriate amount of organoid subculture buffer g to resuspend, and centrifuge for 5min at 300g to discard the liquid (4) add an appropriate amount of organoid cryopreservation solution F, gently blow and resuspend. Take a 24 well cell culture plate as an example: the density is 2 wells and 1 tube is frozen, with a volume of 1.4ml per tube (5) mark the information, and after cooling down the program, transfer it to liquid nitrogen for long-term storage 4. Organoid resuscitation (1) take 10ml of organoid subculture buffer g and put it into a 15ml centrifuge tube (2) take out the frozen organoid cells from the liquid nitrogen tank and quickly put them into a 37 ℃ water bath pot for melting (3) during the water bath thawing process, it is necessary to gently shake the freezing tube to ensure that the frozen solution is completely thawed in 1-2min (4) quickly transfer the dissolved organoid cells to a 15ml centrifuge tube, gently blow 6-8 times with a pipette, centrifuge at 300g for 5min, and then remove the supernatant and collect the organoid cell pellet. Add an appropriate amount of organoid subculture buffer g to resuspend and transfer into a 1.5ml centrifuge tube. Centrifuge for 5min at 300g (5) resuspend Matrigel, lay 25ul Matrigel per well in a 24 well cell culture plate, place in an incubator for 10-15min to form a gel, and add 500ul of mouse normal intestinal organoid culture medium A. |

He was OBSESSED!!!

Our year and a half old lab was excited the moment he saw it come out of the box ! He had many toys that he tore into shreds within a day so this was his first “heavy toy “ that wasn’t easy to tear .
I think he loves the design and feel of it , as he carried this thing EVERYWHERE and even to bed . Him and his sister share toys but this was his , he claimed it lol
Now if it wasn’t for us telling him to stop trying to kill the Dino he could have easily tore into this thing within a couple of days . However since he started chewing gently he became obsessed with the squeaker , and used that as an alarm that he was up every morning in bed 😂
The quality of the product is actually pretty good but it’s not bullet proof, and our smart dog was able to sneakily tear a whole in the nose and some how pull out the stuffing .
All in all I would definitely buy another toy from this company because they are made with quality for dogs , and lasted about three weeks longer than his average toys ! It was his favorite toy 💕

Not indestructible

When I googled “indestructible stuffy dog toys” this popped up. My 9 month old pup loves stuffies but destroys them quickly and eats stuffing if not caught quickly. Well, this indestructible toy is cute but my dog had it torn apart in under 4 minutes so…🤷🏻♀️

Great for Tough chewers!

My dog is a tough chewer and absolutely loves this toy! He was able to get a little stuffing out of the leg so far and it’s been about a week. This might sound like a short amount of time but it usually takes him under 15 mins to unstuff a large toy even if they are advertised for “tough chewers!” This is a very large size toy and would be good for any larger breed. He carries it around the house and constantly keeps his Dino with him. Since Ive found GoDog toys I’ve only been purchasing them because of their durability. I’ve tried Kong and Bark toys previously and they just do hold up long and are usually more expensive. This toy is a great value and dogs love them!

LOVE this brand. Have many of their styles. This is basically an unstuffed toy.

I have about 10 -12 different styles of the GoDog brand toys. It is like Xmas when my Malinois dogs get a new GoDog toy. Have loved this brand for many years and have some toys which have lasted 5-6 years -- minor "surgeries" stitch they back together -- until I retired them. They have never just worn out. Just to be clear:
THESE TOYS ARE NOT CHEW PROOF. NO STUFFED TOY IS CHEW PROOF.
However, this brand of stuffed toy is more chew proof than other brands. But if you are looking for a chew proof stuffed toy be advised -- IT DOES NOT EXIST!! No fabric is chew proof. Even if they invented such a fabric I would not want to ruin my dog's teeth by letting them try to chew through it. Protecting one's dog's teeth should be the highest priority.
If a dog can chew through a PLASTIC KENNEL and even METAL WIRE KENNELS (yes, it happens) then they can easily chew through a puffy, fluffy stuffed toy. The "Chew Guard Technology" language may be misleading to some customers who are disappointed in the brand. I defend the brand and have used it for many years. No, I don't get paid or get free toys.
Since I have had many styles within this brand and even have the pink version of this style I thought I knew what I was getting. This is basically a toy without stuffing which some people prefer. For whatever reasons my dogs prefer toys that are stuffed. Really stuffed with a firm density that they can sink their teeth into. So this toy will be OK. I can tell it will not be one of their beloved toys like some of the brand's other styles such as the purple brontosaurus. My fault for not reading more closely and assuming the grey version would be exactly like the pink version which is fully stuffed. If they get bored with these two I keep some of the Chew Guard toys stock piled.
BTW one can wash and dry these toys. Yes, eventually the squeakers suck up some water and die but one can replace the squeakers. Also on Amazon. So I am willing to replace the squeakers a few times in the life time of one of these toys to be able to wash and dry them occasionally. Usually takes a couple drying cycles to get the heavily stuffed toys dried completely.
If you have an aggressive chewer you may wish to look into Goughnuts dog toys or other hard rubber toys that are designed for aggressive chewers. Even then, with a determined dog, the Goughnuts toys can be chewed through. I think they offer a replacement guarantee. The other brand which has some toys of hard rubber is West Paw. An excellent brand that also offers a one time replacement. Bully Sticks are another option for chewers.
PLEASE, PLEASE, PLEASE stay away from the hard plastic chew toys like the Nyla Bones. Hard plastic toys will destroy your dog's teeth over time. I learned the hard way (no pun intended) and ruined one of my dog's teeth. Vet asked if I allowed my dog to chew ROCKS !!!!!! Every tooth was cracked or fractured. Not wishing to pull teeth I had to pay for some very expensive root canals and caps. Vet's advise: If you can't dent the toy or chew with your finger nail DO NOT offer to your dog. Good advise to follow. Even some of the Bully Sticks (Costco Brand) are so hard they can chip or fracture teeth.
